 Plant development is characterized by plastic, robust, and autonomous morphogenesis and multicellular organization without cell migration and sorting. For elucidating fundamental regulatory systems of cell proliferation and pattern generation underlying these features, we have been studying de novo formation and regeneration of plant organs, which should involve all such systems. The followings are our current major research topics.
シュート再生過程における細胞増殖パターンの制御
Regulation of cell proliferation patterns during shoot regeneration
 シロイヌナズナの組織培養では、高濃度のオーキシンを含む培地で培養してまずカルスを誘導し、次に高濃度のサイトカイニンを含む培地に移植してシュート再生を誘導します。シュート再生の最初期段階では、カルス表層で局所的に細胞分裂が活性化して、不定芽原基のもととなる細胞塊が形成されます。私たちはこのときの細胞分裂に温度依存的な異常を示す温度感受性変異体を複数単離しており、これらを手掛かりに細胞増殖パターンの制御を分子遺伝学的に研究しています。例えば、rid2変異体やrid3変異体の場合、細胞分裂の限局化が不完全で、細胞塊が歪に大きくなり、シュート再生がうまく行きません。rid2rid3とも、プレrRNAのプロセッシングに関与する遺伝子の機能低下が原因であることがわかり、rRNA生合成とシュート再生時の細胞増殖パターンとの予期せぬ関連が浮かび上がってきました。現在は、このように変異体の解析から突き止めた細胞増殖パターンの制御要因を糸口として、その分子機構に迫ろうと研究を進めているところです。また、この課題に関しては、新たにトレニアを用いた研究を始めることも考え、最近になってシュート再生実験系の本格的整備に着手しました。トレニアの茎断片の組織培養では、カルス段階を経由せずに、表皮から直接不定芽原基が生じ、シュートが再生することが報告されているので、この特長を活かした研究を計画中です。
 In tissue culture of Arabidopsis (Arabidopsis thaliana), explants can be induced to form callus on auxin-rich medium and then induced to regenerate shoots by transfer onto cytokinin-rich medium. At a very early stage of shoot regeneration, cell division is locally activated on the callus surface to form pre-meristematic cell mounds, which eventually develop into adventitious buds. We isolated temperature-sensitive mutants that display abnormalities in cell proliferation at this stage of shoot regeneration in a temperature-dependent manner. By molecular genetic analyses of these mutants, we have investigated control of cell division patterns. For example, in the explants of the rid2 and rid3 mutants cultured under a high temperature condition, cell division is not properly restricted, cell mounds are irregularly expanded, and shoot regeneration does not occur. Both rid2 and rid3 have a defect in pre-rRNA processing, which suggests an unexpected relationship between rRNA biosynthesis and cell division control during shoot regeneration. From such clues obtained through analyses of temperature-sensitive mutants, we are approaching molecular mechanisms of the control of cell division patterns. Recently, we have started an attempt for introducing tissue culture of Torenia (Torenia fournieri) into the study of shoot regeneration. Direct shoot regeneration without the intervening callus stage can be induced from the epidermis in tissue culture of stem explants of Torenia. We are planning a new line of research taking advantages of this feature of Torenia tissue culture.
側根原基形成初期過程における不等細胞分裂の制御
Regulation of asymmetric cell division at an initial stage of lateral root primordium formation
 側根原基の形成は、主根内鞘における不等垂層分裂に始まります。この不等分裂によって生み出された小さな細胞の集団が側根原基の土台となるため、不等分裂の回数が側根原基の大きさや形に決定的に重要です。高温条件下でしばしば帯のように幅が広がった(帯化した)側根を形成するシロイヌナズナの変異体を数系統見つけて、これらを詳しく調べたところ、内鞘細胞の不等分裂が余計に起きたせいで帯化側根が生じていることが判明しました。さらに原因遺伝子の解析から、意外にもこの現象にミトコンドリアのmRNAの編集や代謝が関わっていることがわかってきています。これらの新発見から、ミトコンドリアが大きな役割を果たす、未知の細胞分裂制御システムの存在を想定し、その解明を目指して研究を進めています。
 Development of lateral root primordia begins with a few rounds of asymmetric, anticlinal cell division in the pericycle of the primary root. Since a chain of short cells produced by this asymmetric division provides the base of a lateral root primordium, the number of rounds of the asymmetric cell division is critical in the control of its size and shape. We have characterized several temperature-sensitive mutants of Arabidopsis that form broaden (fasciated) lateral roots under high temperature conditions, which attributed the root fasciation in these mutants to by extra rounds of asymmetric division f pericycle cells. Molecular identification of responsible genes for these mutants has revealed unexpectedly that they all are related to mitochondrial mRNA editing/metabolism. We now hypothesize a novel regulatory system of cell division involving mitochondrial functions, which is one of the main targets of our current work.
非標準的葉序の解析による側生器官位置決定機構の再検討
Re-examination of the mechanism determining the location of lateral organogenesis by analysis of noncanonical patterns of phyllotaxis
 葉や花といった側生器官の形成位置がどのように決まるかという問題は、それらの配列様式(葉序、花序)に見られる不思議な数学的規則性も相俟って、古くから多くの人々の興味を掻き立ててきました。近年になって、数理生物学的解析と様々な実験的解析により、側生器官原基が新たな側生器官原基の形成を抑制する作用を周囲に及ぼすこと、この作用がオーキシン極性輸送に依存することなどが明らかにされ、標準的な葉序、花序を一通り再現できる側生器官位置決定の数理モデルも発表されています。しかし、標準から外れた葉序、花序には、これらのモデルでは説明が難しいものもあります。その一つが、開度が180°、90°、180°、270°、180°、・・・というように周期的に変わるコクサギ型葉序です。私たちの研究室では、こうした非標準的な葉序、花序について、数理モデルと実験・観察による解析を行い、側生器官位置決定機構の見直しを進めています。
 It has long attracted interest how the location of lateral organs such as leaves and flowers is determined, since their arrangements, i.e., phyllotaxis and inflorescence, appear to conform to mathematical rules involving mysterious numbers. Recently theoretical studies and experimental studies showed that lateral organ primordia exert a repulsive effect on the formation of lateral organs and that this repulsive effect depends on auxin polar transport. Mathematical models of determination of lateral organ locations based on the repulsive relationship between lateral organ primordia successfully reproduced various canonical patterns that are usual in phyllotaxis and inflorescence. It seems, however, that these models cannot account for some noncanonical patterns of phyllotaxis and inflorescence. A good example of such noncanonical patterns is orixate phyllotaxis, in which the divergence angle periodically changes between three different values, 90°, 180°, and 270°, in the order of 180°, 90°, 180°, 270°, 180°, and so on. We are now re-examining the mechanism determining the location of lateral organogenesis by mathematical modeling and morphological observations focusing on noncanonical patterns of phyllotaxis including the orixate type.
